The handling time you set will be counted towards the Expected Ship by Date and Estimated Delivery Date. Is Sunday considered as a business day towards for Handling Time calculation?

No, Sunday is not counted as a business day and will not have an impact on Expected Ship by Date. For example, if you set 1-day handling time and if you receive an order on Saturday, your Expected Ship by date will be Monday by 11:59pm.

Is Handling Time taken into consideration for Seller Fulfilled Prime and Premium Shipping products?

For Seller Fulfilled Prime (SFP) and Premium Shipping (Next Day Delivery, and Two-Day Delivery), you are always required to ship the product the same day as long as the order is received before the order cut-off time.
